Where do they get these formulas or information

A map square is described in developer coordinates as a 5-tuple of non-negative numbers (z, x1, y1, x2, y2), with the origin in the unexplored ocean to the southwest of RuneScape.

How did they get that information (formulas and such)? Also what do they mean by 64sq by 64sq? Is that the entire RS map or just a portion?

I would assume a map square refers to the entire runescape map (with separate squares referring to floors 1+ in relation to the map). based on:
with the origin in the unexplored ocean to the southwest of RuneScape.

With multiple map blocks per map square, and (x1,y1) referring to an individual map block, while (x2,y2) refer to a point within the given map block, I would assume it's a 64 by 64 tile area.

This would be fairly similar to our SPS map. (x1,y1) decides which of the Runescape_Surface maps, and x2,y2 is the position (or in this case, tile) of the smaller map.

It took me a moment to figure out what this meant, but the map is split into smaller maps 64x64 tiles. If i am remembering correctly, a standard sps map is 500x500 px, and one tile is… 4 px, so a sps map is the same as a 125x125 tile map. Nearly 4x as much area as that of a runescape map. (4096 vs 15625)
To convert the (z,x1,y1,x2,y2) to our 'sps' x and y, it would simply be (x1*64 + x2, y1*64 + y2).

I got confused when you used 64 sq by 64 sq as usually… sq is used as an abbreviation for a square length (area). a 64 tile^2 by 64 tile^2? thats… a 4th dimensional number.

Yeah there's 4096 RS Maps in total. All joined together to make the world map.

:S But a single RS map is 512x512 px :l

So a rs map is then made up of 4 different 'coordinates'. If youre not understanding i can draw something out to explain it better. but i think you got it. :)
